After 3 hours of expression, the cells were fixed and labeled with a polyclonal (a) or monoclonal (c) anti VSV-G antibody and the 100/3 monoclonal anti--adaptin (b) or the polyclonal anti--adaptin (d) antibody, to detect AP-1 and AP-3 complexes, respectively. QNR-71 was detected using FITC-conjugated secondary antibodies. - and -adaptin were decorated with Texas Red-coupled secondary antibodies. Under these experimental conditions, the bulk of the expressed protein is detected in the perinuclear area and has not yet reached the endosomes. Non-transfected cells are indicated with an asterisk.
