Testis was embedded in Unicryl embedding resin and ultrathin sections were reacted with antibodies specific for either usherin (rabbit anti-usherin) or type IV collagen (goat anti-collagen IV). Sections were then reacted with both 10 nm gold-conjugated anti-rabbit secondary antibodies and 5 nm gold-conjugated anti-goat secondary antibodies.
