Control (H1495), sec23-1 (H1496) and sec21-1 (H1497) cells were preincubated at 37°C for 15 minutes, and pulse-labelled with [³⁵S]methionine/cysteine for 5 minutes. Parallel samples were chased with CHX for 30 minutes, as indicated. TM was present from the preincubation onwards, as indicated. The cell lysates were immunoprecipitated with antibody against pentahistidine only (lanes 1-12), or reimmunoprecipitated with antiserum against 1,6-mannose (lanes 13-15), followed by SDS-PAGE analysis.
