Rat-1/tsLA29 fibroblasts (left) and MDCK/tsLA31 cells (right) were seeded on plastic dishes at high density and incubated overnight at 40°C or 34°C. To prevent growth, cells were treated with mitomycin C for 2 hours before band-stripping, then allowed to migrate into the wounded area for 6 hours in DMEM with 10% FCS, fixed and stained. The photographs present typical examples, with horizontal lines, clearly marked on plastic, indicating the initial edge of the wound (top half of each panel). Scale bar: 100 µm. (B) Quantitation. Spontaneous motility was estimated by counting cells that had colonised the margins of stripped areas after 6 hours.
