After 2 days, the cells were incubated under basal or stimulatory (0.3 mM ATP) conditions for 10 minutes. The amount of secreted hGH in the assay medium as well as unsecreted hGH in the cells was determined by ELISA, and the percentage of total hGH secreted was calculated.
