(A) SPARC+/+ and SPARC/ were injected with 107 green-LX-microspheres in the dorsal side of the right ear, which was painted with DNFB 30 minutes later. Draining LNs were collected after 8 and 24 hours and cells were immunolabeled with PE-conjugated anti-I-Ad/I-Ed for two-color FACS analysis. A higher number of green-LX-loaded/I-Ad/I-Ed+ cells were found in draining LNs from SPARC/ than from SPARC+/+ mice at both 8 and 24 hours after treatment. (B) Morphology of green-LX-loaded/I-Ad/I-Ed+ cells in the draining LN reveals the typical hairy and veiled appearance of DCs. (C) Total cell number in the draining LN of treated mice. Migration of green-LX-loaded/I-Ad/I-Ed+ cells in the draining LN is associated with increased LN cellularity which is more extensive in SPARC/ mice.
