Fig. 2. PKC{epsilon}-/- fibroblasts show reduced appearance of {alpha}-SMA stress fibers and `supermature' focal adhesions on fibronectin. PKC{epsilon}+/+ (WT) and PKC{epsilon}-/- fibroblasts cultured on fibronectin were fixed with paraformaldeyde and stained to detect phosphorylated FAK (pFAK), FAK, {alpha}-SMA, total actin, and vinculin. Cells were counterstained with DAPI to detect nuclei (blue stain). PKC{epsilon}+/+, but not PKC{epsilon}-/-, cells show foci of phospho-FAK staining (pFAK) or intense vinculin staining, which are characteristic of `supermature' focal adhesions.
