Recruitment and activation of Rac1 by the formation of E-cadherin-mediated cell-cell adhesion sites

In MDCKII cells transfected with pEGFP-Rac1 wild-type and cultured in normal Ca2+ levels (~1.8 mM), EGFP-Rac1 was localized at sites of cell-cell contact. Whereas, in cells treated with EGTA, EGFP-Rac1 was translocated from sites of cell-cell contact to the cytosol and became diffusely distributed there. After the restoration of Ca2+, EGFP-Rac1 was detected at sites of cell-cell contact. The formation of E-cadherin-mediated cell-cell adhesion sites was completed 120 minutes after the restoration.
