Lens fiber cells from +/+ and SPARC-null (-/-) animals were separated into cortical and nuclear fractions (see Materials and Methods). Proteins were extracted into water-soluble, urea-soluble and pellet (urea-insoluble) fractions and were resolved by SDS-PAGE under reducing conditions.
