QuickTime Video JPEG Image

Movie 1

Figure 1A, 2A: Time lapse imaging of PC-FP expressing Vero cells in the absence of ascorbate. Frames are 1s apart.

Movie 2

Figure 1B, 2B: Time lapse imaging of PC-FP expressing Vero cells 10 minutes after the addition of ascorbate. Frames are 1s apart and the movie loops continuously. Note the movie shows a different cell to the still image emphasising reproducibility.

Movie 3

Figure 2G-J: PC-FP exits the ER in close proximity to COPII labelled ER exit sites. Shown are still images from a time-lapse series showing part of a cell expressing PC-CFP (green) and YFP-SEC24Dp (red). Note the movement of SEC24Dp and PC-CFP together followed by segregation of PC-CFP from YFP-SEC24Dp. The PC-CFP labelled structure passes close by another YFP-SEC24Dp structure before becoming lost amongst the large amount of label in the Golgi region. Images are 4 seconds apart.

Movie 4

Figure 7A: PC-GFP dynamics immediately following injection of anti-EAGE. Cells were injected with anti-EAGE 30 minutes after the addition of ascorbate to the culture medium. Immediately after microinjection of anti-EAGE (in the presence of ascorbate), most PC-GFP labelled structures are immobile, a small proportion (1-5 per cell) remain motile and traffic to the Golgi (frames are 3 seconds apart).

Movie 5

Figure 7B: PC-GFP dynamics 30 minutes after injection of anti-EAGE in the presence of ascorbate. At later time periods after injection (20-30 minutes) few punctate structures remain in the cells but those that do are immobile (frames are 3s apart).
