This movie shows a rotation of the 3D model in Fig. 4. It is a transverse section through a single canaliculus with part of its surface removed to give a clearer view of the many microvilli on the luminal surface.

In this movie, which ultimately contains the 3D mitochondrial model shown in Fig. 5, only the contours of the finished objects are initially shown. The contours are first collected into finished meshed models using IMOD, then the entire mitochondrial model rotates. Interconnections among the mitochondria are readily apparent in the rotation.

Movie 4

This movie shows a stack of 160 sequential tomographic sections penetrating through approximately 0.3 mm of parietal cell interior space. Note the remarkable continuity of the data as individual sections pass through mitochondria, tubulocisternae and the microvilli-lined canalicular region. A sample of a single tomographic section is shown in Fig. 7A.

The 3D model of H,K-ATPase-containing membranes created with a tomogram of resting parietal cells from Fig. 7A is rotated here. A region was selected that shows cisternal, tubular and vesicular elements, although cisternae dominated the overall profile of these membranes (each element is assigned a distinctive color). Note the distinct lack of connections between individual membrane elements.
