Movie 1

Dynamics of neurogenesis: a real time monitoring. For live monitoring of neuro- and gliogenesis, the one week postplated (d7+7) EBs were dissociated and replated. The day after plating the live events were recorded every 30 minutes for three days from the isolated cells. Movie 1 demonstrates the neurogenic progression from a single EGFP+ neural progenitor that is mitotically active. The progenitor divides and one of the daughter cells undergoes differentiation giving rise to a neuron with uni-polar (11.5 hours) and subsequently bipolar (12 hours and after) morphology. Interestingly the other daughter cell still maintains the progenitor potential as evident from its subsequent division at 23 and 36.5 hours. At the same time, the developing neuron migrates away from this progenitor and interacts with neighbouring cells.

Movie 2

Gliogenesis. The proliferating potential of the glia is shown along with their migration and interaction with neighbouring cells. The glial differentiation from the EB-derived cells is observed at a later time point compared with that of the neurons. One of the EGFP+ glial cells undergoes division (55 hours) after transforming from an elongated/spindle shaped morphology to a round one (54.5 hours) resembling the parent progenitor in shape. Subsequently the daughter glial cells become flat, undergo profuse migration and establish interaction with each other and the neighbouring glia (59-66.5 hours).
