Movie 1
GFP-peripherin movements in the growth cone. Time-lapse movie of a growth cone of a GFP-peripherin-expressing PC12 cell plated into DM and observed 4 hours later. Peripherin particles and squiggles are present throughout the growth cone. The GFP fluorescence is viewed with and without phase-contrast images. Images were collected at ~5 second intervals and are played at six frames per second. Images were collected using a Zeiss LSM 510 microscope equipped with a 100¥ oil immersion 1.4 NA lens.
Movie 2
Peripherin particle motility. Time-lapse movies of neurite processes in GFP-peripherin-expressing PC12 cells plated in DM for 4 hours. Particles are observed to be moving both towards and away from the cell body at rates up to ~2 mm/second. The majority of particles in this region (just distal to the cell body) of a neurite are observed to move without pauses or reversals. Images were collected ~5 second intervals and are played at six frames per second. Images were collected using a Zeiss LSM 510 microscope equipped with a 100¥ oil immersion 1.4 NA lens..
Movie 3
Peripherin neurite motility. Photobleach zones were made along the length of neurites to observe peripherin particle and squiggle motility in well-differentiated GFP-peripherin-expressing PC12 cells. During photobleach recovery, many peripherin particles and squiggles were observed to move both into and out of the bleach zone. The movie is played at a speed of six frames per second. Images were collected using a Zeiss LSM 510 microscope equipped with a 100¥ oil immersion 1.4 NA lens.
