Movies 1 and 2. Dynamic movement of Meu14-GFP in live meiotic cells as revealed by time-lapse observations. Cells incubated on a EMM-N plate were suspended in EMM-N medium in the presence of Hoechst33342 and mounted on a glass slide for observation under a microscope controlled by DeltaVision^TM software. Merged images of Meu14-GFP (green) and nuclei (blue) of the cells passing from prometaphase II to metaphase II are recorded at 2 minute intervals (Movie 1). Movie 5. Three-dimensional reconstruction of the Meu14-GFP structure during anaphase II. Cells containing the Meu14-GFP fusion allele were observed under a microscope, z-sections of 0.2 mm were taken and the image was reconstructed. When an enlarged view is rotated on the Y-axis by an angle of 72 degrees, Meu14-GFP rings are found to form a structure like Ôassembled beadsÕ.
