Files in this Data Supplement:
Movie 1. Low magnification (103 objective) time-lapse movie of a control embryo starting at 10 hpf and ending at 12 hpf. PGCs were labeled with GFP by injection of GFP-nos1-3¢UTR RNA. Frames were captured every minute and replayed at 10 frames per second so that each second in the movie represents 10 minutes of real time.
Movie 2. low magnification (103 objective) time-lapse movie of PTX-nos1-3¢UTR injected embryo from 10 hpf to 12 hpf. PGCs were labeled with GFP by co-injection of GFP-nos1-3¢UTR RNA. Frames were captured every minute and replayed at 10 frames per second so that each second in the movie represents 10 minutes of real time.
Movie 3. high magnification (633 objective, digital zoom 2.3) confocal movie of a PGC at 13 hpf expressing PHCrac-EGFP. Frames were captured every 10 seconds and replayed at a speed of 6 frames per second. One second in the movie corresponds to 1 minute of real time (10 minutes in total).
Movie 4. high magnification (633 objective) confocal movie of a migrating PGC labeled with PHCrac-EGFP. Frames were captured every 5 seconds and replayed at a speed of 10 frames per second. One second in the movie corresponds to 50 seconds of real time (5 minutes in total).
Movie 5. high magnification (633 objective) time-lapse movie taken on a non-confocal light microscope of a PGC at 9 hpf expressing PHAkt-EGFP. Frames were captured every 5 seconds and replayed at a speed of 10 frames per second. One second in the movie corresponds to 50 seconds of real time (5 minutes in total)
Movie 6. High magnification (633 objective, digital zoom 2.5) time-lapse confocal movie of a control PGC at 9 hpf. The membrane was labeled using farnesylated EGFP. Frames were captured every 10 seconds and replayed at a speed of 6 frames per second. One second in the movie corresponds to one minute of real time (10 minutes in total)
Movie 7. High magnification (633 objective, digital zoom 2.5) time-lapse confocal movie of a PGC expressing dnPI3K at 9hpf. The membrane was labeled using farnesylated EGFP. Frames were captured every 10 seconds and replayed at a speed of 6 frames per second. One second in the movie corresponds to one minute of real time (10 minutes in total).
