Movies 1-4. v-Src increases spontaneous motility in Dunn chambers. Cells were plated at low density on glass slides and incubated overnight at 40°C or 34°C. Slides were placed in Dunn chambers in medium devoid of serum and growth factors, and transferred on microscopic stage in humidified incubators maintaining temperatures in the wells a 40°C or 34°C±0.1°C, under 5% CO2. Phase-contrast images of cells were recorded using an inverted Olympus microscope coupled to a CCD video camera at 2-minute intervals for 7 hours. Series of images were compressed into movies. In movies 2 and 4 selected motile cells were painted in the Flash5 programme. The last frames of these movies are trajectories of individual cells, reconstructed using ‘Retrac’ software (http://mc11.mcri.ac.uk/retrac/).
