Movie 1. Xin repeats protect stress fibres from LatA-induced disassembly. A7r5 fibroblasts co-expressing mRFP-actin and EGFP-tagged XR1-16 or EGFP alone were subjected to two-colour fluorescence video microscopy before, during and after treatment with 0.5 mM Latrunculin A (LatA) as indicated. Notice the lack of stress-fibre disruption following LatA treatment in the cell co-expressing XR-1-16-EGFP (top) compared with the cell co-expressing EGFP alone (bottom). By contrast, lamellipodia were rapidly disassembled by LatA in both cell types. Movies correspond to a brief time period (3 minutes) before treatment followed by LatA treatment (15 minutes) and recovery period (~23 minutes) as indicated.
