Fig. S1. Myotube striation depends on substrate stiffness. Collagen I-coated polyacrylamide gels were prepared as described elsewhere (Engler et al., 2004a) except that the collagen was micropatterned. C2C12 myoblasts were plated onto the gels or glass substrates and found to fuse into ‘nascent’ myotubes on all. However, after 4 weeks (carried out in triplicate), only the myotubes growing on gels with a tissue-like stiffness differentiated beyond fusion and showed actomyosin striations (Engler et al., 2004b).
