HeLa cells were transfected with either pGEM-U6pHP1, pGEM-U6pHP3 or the control constructs pGEM-U6pcHP and pGEM-U6pT and subsequently arrested by HU treatment as described in Materials and Methods. Samples were prepared for FACS analysis by staining with propidium iodide immediately prior to the release from HU block (0 hours) or 7 hours after release. (A) FACS profiles. The boundaries that define cells with G0/G1, S- and G2/M-phase DNA content were determined as described in Fig. 4A. (B) Cell cycle distribution. Shown is the percentage of cells with G0/G1, S- and G2/M-phase DNA content. Results are representative of two independent experiments.
