  Fig. S1. Untagged dominant-negative mutant does not affect secretory GFP. To facilitate visualisation of secGFP in the apoplast, the Agrobacterium strain was inoculated into the leaf at OD₆₀₀ 0.03 and cells were imaged 48-52 hours post-infection when the rate of secGFP synthesis and transport to the apoplast is at its greatest (I.M., unpublished). Untagged-AtRabF2b was inoculated into the leaf at OD₆₀₀ 0.05. (A) secGFP alone. (B) secGFP and untagged-AtRabF2b-[wt]. (C) secGFP and untagged-AtRabF2b-[S24N]. (C) secGFP and untagged-AtRabF2b-[C198,199S].  The fluorescent pattern and intensity of secGFP is not affected by the co-expression of untagged- AtRabF2b-[wt] or mutants.
