Fragments of PX are embedded in contaminating cell-wall fragments of other cell
types and cytoplasmic material. The GRPCWM (green fluorescence) is associated
with PX. However, some younger PX are not yet associated with GRPCWM (arrows).
(C,D) Cell-wall fraction purified with GRP1.8 antibodies, DSBX-biotin goat anti
rabbit and streptavidin-ferrofluid conjugates. (E,F) Same material as in (C,D)
after extraction with SDS. The cytoplasmic contaminants are removed and the
isolated PX have an increased tendency to stick together. (G,H) Supernatant of
the separation with streptavidin-ferrofluid, containing cell-wall fragments,
cytoplasmic material and protoxylem containing no or little GRP. Less than 10%
