The four videos illustrate the timings of flagellum-flagellum detachment in trypanosome procyclic cells. Trypanosomes are highly motile, in order to view flagellum-flagellum detachment we have filmed live cells that were adjudged to be at a late stage in the cell cycle and that have attached to the slide.

Movie 1. In this video clip the cell body is attached to the slide. The posterior of the cell is on the left of the screen and the anterior end of the cell is on the right of the screen. The cell has two flagella and a cleavage furrow has been initiated from close to the anterior end of the cell. The end of the old flagellum is at the most anterior end of the cell and the new flagellum originates close to the posterior end and its distal tip is attached to the old flagellum close to the anterior end of the cell. Both flagella beat and for a period of time the connection between the tip of the new and the lateral side of the old flagellum is maintained. At around 7 seconds of the video the connection between the new and old flagella is severed.
Movie 2. A cell in late cleavage is observed with its posterior end at the bottom of the frame and the anterior end at the top of the frame. The distal tip of the new flagellum is attached to the lateral aspect of the old flagellum. Both flagella beat and at around 9 seconds of the video the connection between the new and old flagella is severed.
Movie 3. This movie shows two cells in which cleavage is complete but which are still joined by the flagella connector. Both flagella beat and at around 16 seconds of the video the connector attachment is severed.
Movie 4. This movie shows two daughter trypanosomes after the completion of cytokinesis. The daughter cell with the new flagellum (facing the right hand side of the screen) is attached by the connector at the tip of its flagellum to the lateral aspect of the flagellum of the other daughter (which possesses the old flagellum). Flagella of both cells are actively beating and the connector attachment is severed at approximately 10 seconds into the movie.
