  Fig. S1. Lysates obtained from all cell lines used in this study were analysed by western blotting for Pontin, Reptin and Hint1 with anti-Pontin (5G3-11), anti-Reptin (2E9-5) and anti-Hint1 (a-21) antibody, respectively.

  Fig. S2. Functional analysis of shRNAs directed against Hint1 using the pJOsiCheck reporter system. Interference of shRNAs results in a degradation of a transcript encoding luciferase fused to the cDNA of Hint1. In consequence, Renilla luciferase activity is reduced compared to firefly luciferase used for normalization. In these assays shRNA-Hint1_430/431 results in a dose-dependent reduction of Renilla luciferase activity whereas the shRNA-Hint1_432/433 only revealed minor effects.
