Movie 2.
Shear-flow-induced Ax2 motility at 1000 mM external calcium concentration. Shear
stress=2.4 Pa to the right. 30 seconds per image recording. The elapsed time is
given in seconds. 2.53 magnification. The size of the images is 123731010 mm.

Movie 3. Shear-flow-induced
Ax2 motility at low force to illustrate the bi-directional movement observed in
the adapted phase. External calcium concentration=1000 mM. Shear stress=0.5 Pa to the right. 30 seconds
per image recording, total recording 15 minutes. The elapsed time is given in
seconds. 2.53 magnification. The size of the images is 13403965 mm. At the beginning of the recording, cells
move in the direction of the flow. After about 5 minutes, more and more cells
reverse their orientation and move against the flow. During the last 5 minutes,
the cell population is therefore oriented with respect to the flow, but the
direction of this movement is bidirectional. As examples, a few cell tracks are
shown. Cells 1 and 2 show constant orientation in the direction of the flow,
cell 3 moves first in the direction of the flow, then against it, cell 4
initially does not move then starts moving upstream.

Movie 4. Phase contrast
recording of Ax2 undergoing shear-flow-induced motility after raising the
external calcium concentration to 1000 mM. Shear stress=2.4 Pa to the right. 1 second
per image recording. The recording starts 150 seconds after changing the
bathing solution (t=0) and the elapsed time is given in seconds. 603 magnification. The size of the
images is 90356 mm. The top left
cell moves very smoothly over the period t=139-189 seconds. The top right cell
is initially immobile (as many cells are immediately after calcium rise) but
then it starts moving in the direction of the flow. Large round pseudopods are
clearly visible at t=174, 189, 200 and 209 seconds. The bottom cell accelerates
upon calcium rise and exhibits quasi-periodic pseudopods at t=155, 165, 175,
187, 197, 206, 217 and 225 seconds.

of Ax2 undergoing shear-flow-induced motility. 5, 100,
300 mM external calcium concentration,
respectively. Shear stress=2.4 Pa to the right. 1 second per image recording.
603 magnification.
The image sizes are 43323 mm,
55333

Movies 8-10.Three-color model of cell movement.
Blue, gained area; red, lost area; green, steady zone. See Materials and
Methods for details. These videos correspond to the RICM recordings of videos

Movie 11. Fluorescence
recording of Calcium-Green loaded Ax2 undergoing shear-flow-induced motility.
Shear stress=2.4 Pa to the right. 1 second per image recording. The elapsed
time is given in seconds. 603 magnification. The image size is 43325 mm. Two discharges of the contractile vacuole
can be seen during the recording at t=24-27 seconds and t=88-91 seconds.

shear-flow-induced motility, at 1 mM calcium concentration. Shear stress=2.4 Pa
to the right. 1 second per image recording. The elapsed time is given in
seconds and corresponds to the abscissa of Fig. 7A. 603 magnification. The size of the
images is 43325 mm.
