(A) SWE1 or CLN2 mRNA levels in synchronized cell cultures of various strains were determined by Northern blotting, using the SWE1, CLN2, and ACT1 probes. Cells of wild-type (YMM180), pkc1-834 (YMM179) and stt1-1 (YMM196) at early log-phase (OD600 of 0.2~0.3) were synchronized with {alpha}-factor in G1, and resuspended in YPD or YPD plus 100 mM CaCl2. Samples were taken at an interval of 20 minutes after release from arrest. (B) Changes in the relative intensity of the SWE1, CLN2 and ACT1 mRNA. The amount of the SWE1 and CLN2 mRNA in A was normalized to a constant level of ACT1 mRNA. In each case, the maximum value for the first cell cycle was referred to as 1. Time 0 is the point of release from the G1-phase arrest.
