• Supplemental Figure 1 - Fig. S1. The Arabidopsis thaliana AtSCC3 (A) coding sequence  and (B) protein similarities. (A) Schematic representation of the AtSCC3 coding sequence. • Supplemental Figure 2 - Fig. S2. AtSCC3 expression in vegetative cells. Root apex cells labelled with anti-SCC3 serum, either in prophase (A-B) or in interphase (C-J). For each cell, two stainings are provided (with anti-SCC3 in green or with DAPI in blue). G1 cells (C-F), G2 cells (G-J). Data were collected from four independent experiments (representing 368 nuclei). For each experiment, a clear repartition of the root nuclei in two main classes of DAPI fluorescence allowed the identification of nuclei in G1 from nuclei in G2 (K). The comparison of SCC3 staining with DAPI staining showed that cells in G1 as well as cells in G2 are stained with anti-SCC3 serum (L).

  To avoid endoreplication often encountered in plant cells, meristem zones were dissected from primary roots of 7-day-old seedlings. Root apex were spreaded and immunostained according to Armstrong et al., (2002). Nuclei were counterstained with 2 µg/ml DAPI. Total intensity of DAPI and Alexa 488 fluorescence was measured with Open LAB 4.0.2. software.
