cells were left unstimulated or were stimulated for 15 minutes with 20 ng/ml IL-6 or 40 ng/ml purified YFP-IL-6. Phosphorylation of STAT3 was analysed by immunoblotting of cellular lysates using an antibody against tyrosine-phosphorylated STAT3. Equal loading was controlled using a STAT3 antibody. (C) COS-7 cells were transfected with an expression vector encoding gp130-CFP. 48 hours after transfection, confocal images from living cells incubated in a perfusion chamber at 37°C were taken using the YFP and CFP channels of the confocal microscope. Cells were stimulated with YFP-IL-6 (40 ng/ml) and sIL-6R
