When such contractile cells are treated with SMA-FP, {alpha}-SMA incorporation in stress fibers is inhibited (see zoomed-in part of the cell c), resulting in {alpha}-SMA accumulation in RLSs, its disappearance from stress fibers, and subsequently `relaxation' of the cell.
