Fig. S1. WAVE1 and N-WASP are localized to InlB-induced ruffles in WAVE2-knockdown cells. WAVE2-knockdown HeLa cells were stimulated or not with 6 nM InlB, fixed and stained with WAVE1 or N-WASP antibodies and phalloidin-488 to detect F-actin. Notably, stimulated cells diplay large circular dorsal ruffles (arrows). Bar, 10 mm (upper panel ) and 5 mm.
Fig. S2. Ena/VASP recruitment during InlB-mediated ruffling. Vero cells, untreated (resting cells; a,b) or stimulated with 4.5 nM of InlB (+InlB; c-f), stained with anti-VASP antibody (c,d) or transiently transfected with Mena-GFP (e,f) and stained with phalloidin-488. Arrows show recruitment of ENA/VASP proteins at F-actin-rich membrane ruffles. Bar, 5 mm.
Movie 1. Phase-contrast live imaging of entry of Listeria monocytogenes hyperinvasive strain (BUG 1641) (Bierne et al., 2001) into a Vero cell. Images were acquired with a 3CCD video camera (JVC KI-F50), at 24 images per second over 6 minutes.
