Fig. S1: Protein expression levels of the various Rac1 alleles. To examine protein expression levels in hemocytes, UAS-Rac1myc, UAS-Rac1Y40C.myc and UAS-Rac1F37A.myc were crossed to He-Gal4 and hemocytes were bled from wandering third-instar larvae. The hemocytes were stained for Myc expression. Hemocytes from uncrossed transgenic larvae and He-Gal4 were used as controls. Comparing the uncrossed controls to the overexpressing hemocytes, it is evident that all the constructs had a significant increase in the amount of Myc expression. Imagetrak was used to measure fluorescent intensity of at least 100 hemocytes from three different larvae. *, significant difference (Students t-test, P<0.01) compared with the parental UAS and He-Gal4 strains.
Fig. S2: Plasmatocytes recovered from parasitized HmlD-Gal4 control, UAS-BskIR/HmlD-Gal4, or homogygous Rac1J11 loss-of-function larvae and stained with anti-phospho-FAK (red), anti-phospho-tyrosine (green) antibodies, and Alexa-Fluor-350-phalloidin (blue). In the merged pictures shown, overlap of the staining is yellow or orange.
