DCs plated overnight on poly-L-lysine-coated glass coverslips were fixed with 3% paraformaldehyde, permeabilised with 0.05% Triton X-100 and double stained to detect calpain 1 (green) and actin (red) (A) or calpain 2 (green) and actin (red) (B). Arrowheads in A indicate the colocalisation of calpain 1 and actin at the edges of the podosome core at the rear of the podosome cluster. Magnifications of the boxed areas in A and B with calpain staining and double staining are shown on the right. Bars, 10 µm. (C) Western blots of calpain 1 and calpain 2 in total lysates of DCs.
