Fig. S1. Cbs localizes to the nucleus during prophase. During prophase the ER (green) surrounds the nucleus in syncytial embryos. Three 1-μm-thick optical sections starting near the center of the nucleus (top) in the region of the centrosomes (Cnn, center panels and blue in merge) showing concentrated regions of Cbs staining (left panels and red in merge) in multiple sections within the nucleus (arrowheads). The cytoplasmic pool of Cbs is no longer detectable by metaphase.

Three 1-μm-thick optical sections showing the colocalization of Lva (green) and Cbs (red) at DNA (blue) during chromosome congression in a syncytial embryo. Cbs and Lva are only detectable in sections that contain DNA (arrowheads indicate position of nucleus). Lva is no longer detectable by the time chromosomes are aligned at the metaphase plate.
