Movie 1. GBF1 accumulates on membranes of the Golgi complex and peripheral VTCs upon treatment with BFA. NRK cells expressing GFP-GBF1 were imaged on the temperature-controlled stage of a Zeiss LSM510 confocal laser scanning microscope at 37°C. Single slice images were captures every 2 seconds for 10 minutes immediately after BFA addition (1 μg/ml). The movie displays 15 frames per second.
Movie 2. Nocodazole treatment stabilizes GBF1-positive peripheral VTCs. NRK cells expressing GFP-GBF1 were treated on ice for 15 minutes with either 20 μg/ml nocodazole or 0.5% DMSO. Following a 2 minute warm-up to 37°C, live cells were examined on the heated stage of a Zeiss Axiovert 200M spinning disk microscope. Z-stacks of six slices each 1 μm thick were acquired continuously both before (about 45 seconds) and after (about 10 minutes) addition of BFA (5 μg/ml). Movies display projected stacks that include a time stamp.
