Fig S1. Effect of RIIα depletion on intracellular transport of VSV-G. HeLa cells were incubated (+) or not (−) for 80 hours with a siRNA designated to inhibit the synthesis of RIIα. Cells were infected with the ts045 mutant form of vesicular stomatitis virus (VSV) and incubated at the restricted temperature (40°C) for 4 hours to allow accumulation in the ER of newly synthesized G glycoprotein (VSV-G). Cells were then shifted to the permissive temperature (32°C) for 1 hour to induce VSV-G export from the ER. Presence of VSV-G at the cell surface was detected by incubating the fixed, unpermeabilized cells with an antibody against the luminal domain of this protein. Cells were then permeabilized and stained with antibody against RIIα. Bar, 10 μm.
