Fig. S1. Non-homologous plasmid genes form separate but adjacent transcription domains. (A) Plasmids pHIV-β and pHIV-ε, which are identical except for the β-globin and ε-globin gene sequences, were co-transfected into HeLa cells and the nascent transcripts were detected by RNA in situ hybridisation with probes of the first intron. The plasmids formed separate but adjacent TDs. (B) Plasmids pHIV-α and pHIV-β were transfected sequentially into HeLa cells with a 24-hour interval between the transfections. The nascent transcripts were labelled by RNA in situ hybridisation with probes of the first intron. The plasmids formed separate but adjacent transcription domains indicating that this pattern is not an artefact of the co-transfection protocol. Bars, 5 μm.
