Movie 1. Laser scanning confocal microscopy (LSCM) movies of cells expressing SBDS-GFP demonstrate that SBDS protein is distributed evenly throughout the cytoplasm in cells migrating in buffer (panel A), but enriches in the anterior pseudopod in cells migrating in a spatial gradient of cAMP (panel B). The source of chemoattractant is to the right in panel B. Images were acquired 1 mm above the substratum at 5-second intervals, as described in Material and Methods and in the legend to Figure 3.
Movie 2. Laser scanning confocal microscopy (LSCM) movies of a cell expressing SBDS-GFP while migrating in buffer (panel A) and a cell expressing SBDS-GFP making a lateral pseudopod while chemotaxing in a spatial gradient of cAMP (panel B). In the presence of cAMP (panel B) SBDS-GFP localizes to the site of lateral pseudopod formation very close to the time the pseudopod emerges, remains localized during its expansion, and disappears during retraction. The source of chemoattractant is to the right in panel B. Images were acquired 1 mm above the substratum at 5-second intervals, as described in Material and Methods and in the legend to Figure 4.
