Tor2 inactivation induces rum1+ and ste9+ gene transcription. tor2-51 rum1-d3 (Daga et al., 2003) and tor2-51 ste9-d3 (Alvarez et al., 2006) cells, expressing mutants of rum1+ and ste9+ lacking the 3UTR region that causes mRNA instability, were grown in EMM to the mid-exponential phase at 25°C, shifted to the restrictive temperature of 32°C, and samples were taken at the indicated times. RNA was extracted and northern blots performed using probes against rum1+ and ste9+, respectively.
