Fig. S1. Uncoupling between mid1-4GFP distribution and nuclear position in pom1Δ cells. Wild-type (AP1442; a-b) and and pom1Δ cells (AP 1502, c-d) pretreated with 25 μg/ml MBC were ultracentrifuged as described by Daga and Chang (Daga and Chang, 2005) to move the nucleus away from the cell center and maintained in liquid medium containing MBC at 25°C after ultracentrifugation. Cells were imaged shortly after centrifugation (10-20 minutes after centrifugation; a,c) or 2 hours to 2.25 hours after centrifugation (b,d). Note that mid1p cortical spots are still in the medial region in wild-type cells 10 minutes after centrifugation but relocate to the new cortical region facing the nucleus upon prolonged incubation in presence of MBC (arrowheads). In pom1Δ cells nuclei are either centrifuged towards the non-growing tip containing Mid1p cortical spots (stars in c) or towards the opposite tip (arrows in c). Cells with Mid1p spots distributed in the region opposite to the nucleus (arrows in d) are still observed more than two hours after centrifugation. Bar, 5 μm.
