Movie 1. Overlay of images of Mag-Fura-2 (red) and FM 1-43 (green). Note that mobile ER vesicles were observed mostly in neural processes and the soma was preferentially occupied by the immobile reticulum. The vesicles showed saltatory motion consisting of continuous directed movements (mediated by molecular motors) and pauses (‘wiggling’) corresponded to the confined diffusion (the restricted Brownian motion). Note also that the vesicles decreased their velocity in the vicinity of some synapses (indicated by arrows, but there are many other similar locations). The stops of particles also correlated with the synaptic activity (seen as green blinking). The neurons cultured from the pre-Bötzinger complex (the respiratory network in the brain stem) were stained with TMRE, Mag-Fura-2 and FM 1-43, which label mitochondria, ER and synaptic vesicles, respectively. The images were taken every 5 seconds (acquisition time, 0.3 seconds). The width of both frames=120 μm.
