  Fig. S1. Localization of dsc3 in E6.5 mouse embryos. In situ hybridizations (A,B) demonstrated similar expression patterns of dsc3 (A) and dsc2 (B). Note that trophoblast cells express dsc3 (arrows in A). Dsc2 and dsc3 sense control probes did not yield signals (data not shown). EPC: ectoplacental cone; PAC or *: proamniotic cavity. Immunofluorescence microscopy (C-H) using cryo-sections of E6.5 embryos. Dsc3 staining is shown in red while control antibody staining is shown in green. Overlap resulted in yellow fluorescence. The following antibody combinations were used: Dsc3 + Dsg1/2 (C), Dsc3 + TROMA-I (D), Dsc3 + Dp (E-H). Dsc3 is expressed in the maternal decidua (DC) and various embryonic tissues, in particular the ectoplacental cone (EPC in E-H) and the embryonic ectoderm surrounding the proamniotic cavity (PAC or * in C, D, F, H). Note that Dsc3 partially overlaps with Dsg1/2, desmoplakin (Dp) and endodermal keratins (TROMA-I). Control staining with normal guinea pig and normal mouse IgG, respectively, did not yield a specific staining pattern (data not shown).
