All panels show overlays of single-cell calibrated fura-2 (black trace) and Epac-1 signals (coloured traces) during imposed Ca2+ changes. Cells were pre-treated with Tg (100 nM) in Ca2+-free saline to deplete Ca2+ stores, and PGE1 (10 nM) was added to stimulate basal AC8 activity. Real-time cAMP and Ca2+ changes were obtained at various frequencies and durations. (A) Periodic CCE-evoked Ca2+ entry (2 minutes), and washes in Ca2+-free saline (2 minutes).
