Figure S1. PTEN is required for Sema3A mediated Akt/GSK-3 signaling in breast carcinoma cell lines. (A) MDA-MB 231 or (B) MDA-MB 468 cells were labelled with phalloidin (left) and anti-PTEN antibody (middle). Confocal micrographs, taken with identical scanning parameters, demonstrate the absence of PTEN signal in MDA-MB 468 cells. (C) MDA-MB 231 or (D-F) MDA-MB 468 cells were transfected with the indicated constructs and treated for the specified time with Sema3A (1 mg/ml) for the indicated time (minutes) or LY294002 (10 mM, 30 minutes). Equal protein quantities (10 mg) were subjected to SDS-PAGE and transferred to nitrocellulose membrane, before probing with P(Ser473)Akt or P(Ser9)GSK-3b. Western blotting using anti-PTEN antibody verified the absence of PTEN in untransfected MDA-MB 468 cells (D).
