Serum-starved Swiss 3T3 fibroblasts do not polarize under shear stimuli. (A)
Immunofluorescence micrographs of serum-starved Swiss 3T3 fibroblasts before
and after shear (microtubules, red; nuclear DNA, blue; actin, green). Bar, 20 μm. (B) Polarization assay was conducted as described previously for serum-starved Swiss 3T3 fibroblasts (Lee et al., 2005). Serum-starved cells, either unsheared (n=61) or
sheared (n=80), displayed
isotropic centrosome location with respect to the nucleus (approximately
50:50). Cells cultured in complete media containing 10% BCS exhibited polarized
location of the centrosome with respect to the nucleus when sheared
(approximately 75:25).
