Fig. S1. Acetylated tubulin identifies central microtubules in control and nocodazole-treated cells. MCF-7 cells were fixed after incubation in buffer (Control) or with nocodazole (100 μM, 90 minutes) and immunostained for E-cadherin or acetylated tubulin.
Fig. S2. CLIP-170 lacking the tail domain (CLIP-170 Δ-Tail) disrupts the characteristic radial microtubule pattern in MCF-7 cell monolayers. MCF-7 cells were transiently transfected with CLIP-170 Δ-Tail, then stained for the epitope tag and either β-tubulin (A,A’) or for the epitope tag and acetylated tubulin (B). A higher magnification view of the boxed region is shown in A’. Microtubules in cells expressing the transgene (A’, arrow) characteristically failed to extend into cadherin-based cell-cell contacts (outlined), in contrast to untransfected neighbour cells (A’, arrowhead).
