treatment inhibits protein arginine methylation. (A) Total cellular proteins,
isolated from HeLa cells treated with 0, 10 or 30 μM AdOx for 24 hours, were resolved by SDS-PAGE and probed with pAbs that detect aDMA (ASYM24) or sDMA (SYM11). The aDMA blot was subsequently re-probed with anti-Hsp70cp mAb. The Coomassie-stained gel shows the protein loads for each sample. (B) AdOx inhibits the protein arginine methylation that is recognized by the anti-mRG pAb. HeLa cells treated with 0 or 20 μM AdOx for 24 hours were immunostained with anti-mRG pAb. The anti-mRG immunofluorescence intensity of individual cells was determined using the histogram function in Adobe PhotoShop. The intensity values for three independent experiments, a total of 75 cells under each condition, are plotted. The reduction in anti-mRG fluorescence intensity in the presence of AdOx was significantly different than in its absence (P<0.0004, ANOVA).
