Fig. S1. Immunolocalisation of WT L-plastin and vimentin in Vero cells.Cells were transfected with cDNA encoding WT L-plastin. After 48 hours, cells were processed for immunofluorescence double staining with anti-sv-tag antibody (left panel) and anti-vimentin antibody (middle panel). Alexa-Fluor-488-coupled anti-rabbit IgG (sv-tag) and Texas-Red-coupled anti-mouse IgG antibody served as secondary antibody. Right panel shows a merge of left and middle images. Fig. S2. Binding of S/A L-plastin to F-actin. Six μM of β actin were co-polymerised over night at 4°C with various concentrations of S/A L-plastin and centrifuged at high speed for 30 minutes. Supernatants and pellets were analysed by SDS-PAGE. Coomassie-staining patterns of pellets (top panel) and supernatants (bottom panel) of duplicate samples are shown. The molar ratios of L-plastin to actin are indicated.
