Movie 1. EGF stimulation induces ruffling of E-cadherin-GFP on free cell surfaces. MCF-7 cells stably overexpressing GFP-tagged E-cadherin (hE-GFP-MCF-7 cells) were imaged every 12 seconds over a 1-hour treatment period with EGF using a TILLvision live imaging microscope.
Movie 2. EGF stimulation induces macropinocytosis of E-cadherin-GFP. Arrows in the initial frames of the image sequence indicate an E-cadherin-positive membrane ruffle that is internalized to form a macropinosome (circle and subsequent arrows) in a hE-GFP-MCF-7 cell stimulated with EGF. The movie shows a merged overhead view of three axial slices taken every 5 seconds and was imaged using 4D confocal microscopy (Lock et al., 2005) on a Zeiss LSM510 confocal laser microscope.
