Movie 1. Wild-type pupa undergoing morphogenesis. Movie is captured in real time using an analog Olympus OLY-105 video camera attached to an Olympus SZX12 dissecting microscope. Recording began at approximately 8 hours after puparium formation, and the video is sped-up 60×. At the beginning of the recording period, the prepupa is undergoing coordinated muscle contractions to move air from the posterior end to the anterior, which later dictates adult body position within the pupal case. Several muscle-driven events occur during the recording period: (1) at ∼9 seconds, the larval head retracts; (2) at ∼27 seconds, the larval head involutes; and (3) at ∼33 seconds, the adult head begins to evert.
Movie 2. mlp84B−/− (Df(3R)dsx2M/Df(3R)Scx2) mutant pupa. Movie is captured in real time using an analog Olympus OLY-105 video camera attached to an Olympus SZX12 dissecting microscope. Recording began at approximately 8 hours after puparium formation, and the video is sped-up 60×. This pupa is approximately the same age and developmental stage as the wild-type pupa, based on both timing and morphological features of the pupae. The mutant exhibited only feeble posterior muscle contractions that do not produce enough force to translocate the posterior air bubble. Over a 10-hour period this pupa did not show any signs of larval head retraction or adult head eversion.
