The E95S mutant of the plectin actin-binding domain (ABD) has a reduced affinity for nesprin-3α COS7 cells were transiently transfected with a construct encoding VSV-nesprin-3α (lanes 1-3), VSV-nesprin-3β (lanes 4-6), HA-plectin ABD (lanes 1, 4 and 7) or HA-plectin ABD^E95S (lanes 2 and 5). The cells were lysed in RIPA buffer and HA precipitates were probed for nesprin-3 (upper panel) and HA (second panel). WCLs were probed for the expression level of nesprin-3 (lower panel).
