Mesangial cells serum-deprived for 24 hours were treated with TGF-β1 for 30 minutes, then fixed and stained for FAKs green; phospho-Y397-FAK (top panels), phospho-Y925-FAK (middle panels) or pan FAK (bottom panels), along with vinculin (red, middle panels). Merged images (left panels) demonstrate that phosphorylated FAK mostly localizes to the sites of FA formation, as depicted by vinculin staining.
Fig. S2. HKC cells were transfected with either a wild-type or a Y397F FAK construct along with a GFP expression vector (Clontech Laboratories, Palo Alto, CA). After 20 hours, the cells were fixed and stained for vinculin (red) to detect FA formation. Nuclei are shown as blue with DAPI staining and the cells expressing FAK construct appear green by co-expression of GFP.
